Hi Ab,
Please look at the Tutorial sessions for an
explanation on the concept of regions in
ITK images.

in particular look at

For each of the three types of image regions,
(LargestPossible, Buffered, Requested) you can
only define one region of each type at a time.
The purpose of the regions is to support streaming
and multi-threading on the ITK pipeline.
If what you want is to simply extract a region
from an image you should look at the filters:
- RegionOfInterestImageFilter

- ExtractImageFilter
http://www.itk.org/Insight/Doxygen/html/classitk_1_1ExtractImageFilter.html
You will find examples on the use of these filters
