No. On the contrary, for a 256x256x256 volume registration rigid 
registration with fairly good quality should usually take 5-20 minutes.

Did you plot a trace of the optimizer.

Have you tried the GUI application in InsightApplications ?  
LandmarkInitializedMutualInformation ? It does rigid registration with 
the Mattes metric.
